RESUMO

Objective: To investigate the mechanism of S100A7 inducing the migration and invasion in cervical cancers. Methods: Tissue samples of 5 cases of cervical squamous cell carcinoma and 3 cases of adenocarcinoma were collected from May 2007 to December 2007 in the Department of Gynecology of the Affiliated Hospital of Qingdao University. Immunohistochemistry was performed to evaluate the expression of S100A7 in cervical carcinoma tissues. S100A7-overexpressing HeLa and C33A cells were established with lentiviral systems as the experimental group. Immunofluorescence assay was performed to observe the cell morphology. Transwell assay was taken to detect the effect of S100A7-overexpression on the migration and invasion of cervical cancer cells. Reverse transcription-quantitative real-time polymerase chain reaction (RT-qPCR) was used to examine the mRNA expressions of E-cadherin, N-cadherin, vimentin and fibronectin. The expression of extracellular S100A7 in conditioned medium of cervical cancer cell was detected by western blot. Conditioned medium was added into Transwell lower compartment to detect cell motility. Exosomes were isolated and extracted from the culture supernatant of cervical cancer cell, the expressions of S100A7, CD81 and TSG101 were detected by western blot. Transwell assay was taken to detect the effect of exosomes on the migration and invasion of cervical cancer cells. Results: S100A7 expression was positively expressed in cervical squamous carcinoma and negative expression in adenocarcinoma. Stable S100A7-overexpressing HeLa and C33A cells were successfully constructed. C33A cells in the experimental group were spindle shaped while those in the control group tended to be polygonal epithelioid cells. The number of S100A7-overexpressed HeLa cells passing through the Transwell membrane assay was increased significantly in migration and invasion assay (152.00±39.22 vs 105.13±15.75, P<0.05; 115.38±34.57 vs 79.50±13.68, P<0.05). RT-qPCR indicated that the mRNA expressions of E-cadherin in S100A7-overexpressed HeLa and C33A cells decreased (P<0.05) while the mRNA expressions of N-cadherin and fibronectin in HeLa cells and fibronectin in C33A cells increased (P<0.05). Western blot showed that extracellular S100A7 was detected in culture supernatant of cervical cancer cells. HeLa cells of the experimental group passing through transwell membrane in migration and invasion assays were increased significantly (192.60±24.41 vs 98.80±47.24, P<0.05; 105.40±27.38 vs 84.50±13.51, P<0.05) when the conditional medium was added into the lower compartment of Transwell. Exosomes from C33A cell culture supernatant were extracted successfully, and S100A7 expression was positive. The number of transmembrane C33A cells incubated with exosomes extracted from cells of the experimental group was increased significantly (251.00±49.82 vs 143.00±30.85, P<0.05; 524.60±52.74 vs 389.00±63.23, P<0.05). Conclusion: S100A7 may promote the migration and invasion of cervical cancer cells by epithelial-mesenchymal transition and exosome secretion.

@#【Objective】To investigate the expression of FosB in invasive breast cancer and its correlation to the ultrasound findings and clinical significance.【Methods】Immunohistochemical and ultrasound examination were performed for 155 cases with invasive breast cancer,36 cases with paracancerous tissue and 30 cases with normal breast tissue. To investigate the correlations between the different expression of FosB with histological grade ,lymph nodes,and ultrasound findings.【Results】①The expression of FosB in paracancerous tissue and normal breast tissue was higher than that in invasive breast cancer(P<0.01). ②Based on immunohistochemical staining,high positive expression rate of FosB was associated with low histological grade,ER positive and PR positive(P<0.05). There was no correlation between FosB and age,tumor diameter,lymph nodes,Her2 and clinical stages(P>0.05)③The expression of FosB was higher in microcalcification positive and lymph nodes bigger tissues(P<0.05). There was no correlation between FosB and tumor size, regular shape,boundary,posterior attenuation ratio and blood flow(P>0.05).【Conclusion】The expression of FosB in invasive breast cancer was lower than that in paracancerous tissue and normal breast tissue. The different expression of FosB was correlated with histological grade,ER positive and PR positive. To study the expression of FosB in invasive breast cancer may be helpful for differential diagnosis and targeted treatment. The different expression of FosB was correlated with microcalcification,lymph nodes,and combined FosB with ultrasound findings may contribute to prognostic evaluation of breast cancer.

Objective To investigate the breeding conditions of mosquitoes and larvaes in 12 schools in Xuhui District of Shanghai before and after the beginning of summer term, and to provide basis for mosquitoes control in schools. Methods We used stratified random sampling to select 4 schools from each of the categories: kindergartens, primary and secondary schools, colleges and technical secondary schools in Xuhui District in August and September 2019.We used the methods of human landing catch, visual evaluation and ladling to investigate the breeding conditions of mosquitoes and larvaes before and after the beginning of summer semester. Results The mosquito density was 10.50 (per person per time) after the beginning of term, lower than 19.00 (per person per time) before schools opening, and the difference was statistically significant (χ2=4.20, P < 0.05).The positive rate of larvae breeding in small-scale ponds after the beginning of term was 31.47%, lower than 44.23% before the schools opening, and the difference was statistically significant (χ2=4.22, P < 0.05).The positive rate of larval breeding in medium and large static water was 14.00% after the beginning of term, lower than 17.86% before the schools opening, and the difference was no significant (χ2=1.03, P=0.31). Conclusion Schools need to take the following actions: to focus on the work to manage mosquito breeding sites on campus before the beginning of the term; to strengthen the daily work of environment control; to establish a model of prevention and control in cooperation with multi-medical and educational departments, and to form a collaboration system in application of drugs to rain-water catch basin sand in other mosquito control work in residential areas.

<p><b>OBJECTIVE</b>To evaluate the prognostic significance of preoperative serum albumin in patients with gastric cancer undergoing radical resection.</p><p><b>METHODS</b>A total of 146 patients with gastric cancer underwent radical resection from January 2001 to December 2003. Clinicopathological data were analyzed retrospectively. Patients were divided into two groups, including patients with a normal preoperative serum albumin level(>35 g/L, n=115) and patients with hypoalbuminemia (≤35 g/L, n=31).</p><p><b>RESULTS</b>Patients with a low albumin level were associated with a higher postoperative recurrence rate(90.3% vs. 43.5%, P<0.01). The overall 5-year survival rate in patients with a normal serum albumin level was significantly higher than that in patients with a low serum level(57.4% vs. 9.7%, P<0.01). On multivariate analysis, preoperative serum albumin level was an independent factor associated with survival(P<0.01). When stratified by nodal metastasis, normal serum albumin level was still associated with higher survival rate(P<0.05). Prognostic significance was found in patients with lower stomach cancer(P<0.01), but not in patients with cancer in the upper and middle stomach(P>0.05).</p><p><b>CONCLUSION</b>Hypoalbuminemia is associated with worse survival in patients with cancer in the lower stomach and adjuvant therapy should be considered.</p>

<p><b>OBJECTIVE</b>To analyze the differences among gastric cancers in different locations (upper, middle, and lower) in the clinicopathological characteristics and prognosis of gastric cancer.</p><p><b>METHOD</b>Clinicopathological data of 146 patients with gastric cancer undergone radical gastrectomy from January 2001 to December 2003 were analyzed retrospectively. These patients were divided into three groups:tumors in the upper third of the stomach (group U), middle third (group M) and lower third (group L).</p><p><b>RESULTS</b>There were significant differences in the the Borrmann types among three groups and between every two groups (P<0.01). Compared with group M and group L, cases in group U were older (P=0.034), had a larger tumor size (P=0.001) and limited extent of lymph node dissection (P=0.009). Compared with group M and group U, cases in group L had earlier nodal status P=0.007) and TNM stage (P=0.020), and the recurrence rate was lower (P=0.006). Survival time of group M and group U was significantly shorter than that of group L (P=0.007, P=0.001).</p><p><b>CONCLUSIONS</b>Tumors in the upper third and the lower third of stomach are associated with distinct clinicopathological characteristics whereas tumor in the middle third shares some characteristics of both entities. The prognosis differs significantly among tumor locations.</p>

<p><b>OBJECTIVE</b>To investigate the pattern of solitary lymph node metastasis in order to offer more suitable treatment for patients with gastric cancer.</p><p><b>METHODS</b>Sixty-five patients received operation between July 1999 and June 2004 with only 1 metastatic lymph node identified by postoperative pathological examination were included in the study. Data were analyzed using the statistical software SPSS 13.0.</p><p><b>RESULTS</b>Univariable analysis showed that only the tumor diameter differed significantly between patients with skipping metastatic (SM) lymph node and those without SM (chi(2)=4.447, P=0.035). No clinicopathological factors showed statistically differences between patients with lymph node transverse metastasis (TM) and those without TM. However, both of two comparative groups showed statistically differences in long-term survival (P=0.000, P=0.000).</p><p><b>CONCLUSIONS</b>Most lymph node metastasis in gastric cancer follows the rule of "near-to-far", but some special metastasis patterns (SM, TM) are not rare. Proper lymph node dissection especially for patients with high risk should be performed to reduce tumor recurrence and improve long-time survival.</p>

<p><b>OBJECTIVE</b>To explore the pattern of lymph node metastasis(LNM) in advanced proximal gastric cancer in order to guide lymphadenectomy.</p><p><b>METHODS</b>Between September 2001 and December 2005, a total of 103 patients with advanced proximal gastric cancer underwent radical gastrectomy with D2 or>D2 lymphadenectomy. The clinical characteristics, pathologic features, and LNM were analyzed by univariate and multivariate analysis.</p><p><b>RESULTS</b>LNM was observed in 81 of 103 cases(78.6%). The LNM was identified in 70.8% at N1, 38.3% at N2, 22.3% at N3. LNM frequency was found in groups No.3,No.1,No.2 and No.4Sa,4Sb (from the highest to the lowest) at N1, groups No.7, No.10, No.9, No.11, No.8a and No.4d at N2, and groups No.5, No.6, No.16 and No.12 at N3. Ordinal Logistic regression analysis showed that histopathological type, tumor size, depth of invasion, and distant metastasis were independent factors for lymph node metastasis in advanced proximal gastric cancer.</p><p><b>CONCLUSIONS</b>The number of lymph node metastasis in advanced proximal gastric cancer is mainly associated with differentiation, tumor size, depth of invasion, and distant metastasis. It is essential to dissect the lymph nodes according to the risk of lymph node metastasis.</p>
